The only thing that can be done at the Fordow site is to block the exits. The GBU-57 could be used for that. I don't know if this new site that has been discovered is above or below ground. I don't know anything about this, but it seems like this MOP is merely a better version of previous bunker busters, but the article you posted seems pretty uncertain about it's usefulness against hardened underground sites, noting that some sites are very far underground and/or comman centers are decentralized.

If you build a facility inside a mountain, I'd imagine even nukes wouldn't penetrate.

Bunker busters are usually used in clusters for large hardened targets. I obviously can’t say for certain, but there is an understanding among military brass that number of well placed GBU 57s would likely do the job. You don’t need to obliterate the facility, you just need to make it inoperable and unsafe to be inside - which isn’t terribly difficult to do for an underground facility which uses highly radioactive materials and is entirely dependent on powered ventilation systems.
