Are there any long term negative effects?
I'm not aware of any long term physical side effects - I personally haven't had any (that I'm aware of) nor have I read anything in the scientific literature or personal usage accounts I'm familiar with. The real risks, as others mentioned, are largely mental. The biggest one is that if you are at risk for mental illnesses like schizophrenia, you probably shouldn't be doing them as they can trigger a psychotic episode with potentially lasting consequences. Another big risk is doing something that can cause physical harm while under the influence (walking out in traffic etc), but that is easily mitigated by having a sitter. Depending on the content of the experience and your ability to integrate it, you can also experience depression, existential dread etc, after a trip, but in my experience and from what I've heard from others, mushrooms tend to go gently on you your first few times, if you're taking them seriously. This means no taking them and going to a party or concert - you take them as part of a ritual, offer thanks to them, and then lay down with your eyes closed for the first half of the trip or so. After you can go out in nature a bit, but again, they're not a party drug and if you treat them that way you'll get burned eventually. They may show you things you struggle to deal with, and one of the biggest causes of bad trips is fighting against the experience, but if you let go and surrender to the mushroom things will generally go much smoother. Having an established meditation practice helps immensely here, especially during the initial come up. If you're serious about trying them, take the time to grow them yourself (its really easy and actually pretty fun), and as you're growing them use the time to learn about the experience, begin meditating, figure out if you have any major unresolved issues you're not addressing, etc.
