Most recently, Lyoto Machida attempted to win the Middleweight belt, having already held the Light Heavyweight belt. Not sure why no one else has remembered this.

Edit: Not sure if we're talking about challenging while being champ in another weight class. If so, only BJ Penn. Couture has had belts for Light Heavyweight and Heavyweight, but never at the same time.
